Growth hormone studies: ipamorelin, CJC-1295, tesamorelin
A comparison begins with the population and the outcome. The compound name alone leaves too much unsaid.
Human trials beside each other
Ranking these compounds by desirability is an exercise I distrust. The human studies pursued different ambitions, so the useful comparison has to preserve the question each one asked.
| Compound | People studied | Measured result |
|---|---|---|
| Ipamorelin | 114 adults undergoing bowel resection | First tolerated meal: 25.3 hours versus 32.6 hours with placebo, p=0.15; primary endpoint missed |
| CJC-1295 | Healthy adults aged 21 to 61 | Mean growth hormone rose two- to ten-fold for six days or more; IGF-1 rose one-and-a-half- to three-fold for nine to eleven days |
| Tesamorelin | Adults with HIV-associated lipodystrophy in five pooled trials | Visceral fat mean difference -27.71 square centimetres; 95% CI -38.37 to -17.06, p<0.001 |
Tesamorelin has the approved indication: excess abdominal fat in adults with HIV-associated lipodystrophy. Ipamorelin and CJC-1295 have no approved human indication. There is no head-to-head trial comparing any pair of these three compounds, and no study in this list enrolled healthy trained adults to measure strength or hypertrophy.
CJC-1295 growth hormone pulsatility
Growth hormone normally arrives in bursts. In the CJC-1295 pulsatility study in healthy men, pulse frequency and magnitude were unaltered while trough and mean concentrations rose. The long-acting form's estimated half-life was 5.8 to 8.1 days. A preserved pulse pattern can coexist with substantially prolonged exposure.
Ipamorelin's intravenous pharmacology in healthy male volunteers showed a terminal half-life of approximately 2 hours and a single growth hormone response peaking at about 0.67 hours. Tesamorelin's healthy-men study examined endogenous secretion and insulin sensitivity together, giving it a metabolic observation that the others' hormone measurements cannot supply.
Pulsatility's reassuring sound troubles me. It describes a pattern. Blood sugar, fluid retention, tumour-related concerns and longer-term health outcomes require additional evidence.
Ipamorelin and CJC-1295 together
Ipamorelin acts at the ghrelin receptor; CJC-1295 and tesamorelin act at the GHRH receptor. These are distinct inputs to the pituitary's growth-hormone-producing cells. Their complementary signalling explains the interest in combining ipamorelin with CJC-1295, but the pairing has no controlled human outcome trial.
The GHRH-analogue review supplies pharmacological context. Separate experiments with the individual compounds remain separate experiments. The combination claim exhausts my confidence at the point of testing: neither half has tested it.
Community accounts of sleep, workout recovery and appearance are anecdotal and confounded by diet, training, co-use and uncertain product identity. No amount or schedule can be justified from those accounts, and none is offered here.
Animal weight and human fat
The ferrets receiving cisplatin lost less weight with ipamorelin. GHRH-knockout mice regained normal weight and length with CJC-1295. In human tesamorelin trials for HIV-associated lipodystrophy, visceral and trunk fat declined while lean mass increased.
My table has room for all those experiments. Combining them into one promise about muscle or fat loss would erase the reason for each result. Preserving weight under chemotherapy, restoring deficient growth and reducing a specific fat depot are three different aims.
The individual pages follow those aims further: ipamorelin's meal and animal studies, CJC-1295's human blood measurements and tesamorelin's tissue outcomes.
